Description
iPS-Derived Endothelial Cells Differentiated Media designed for the differentiation of iPS into endothelial cells. Medium contains growth factors and supplements. It is formulated for use with 5% CO2 and 95% air in a humidified incubator. The medium consists of 500 ml of basal medium (containing essential and non-essential amino acids, vitamins, organic and inorganic compounds, hormones, growth factors, trace minerals), supplemented with cell growth supplement, antibiotics, and fetal bovine serum.
Cell Culture Medium and Supplements include:
- 125 ML Ham's F-12 Nutrient Mix
- 375 ML IMDM
- 2.5 ML N2-SUPPLEMENT
- 5.0 ML B-27 Supplement Minus
- 2.5 ML 10%BSA
- 0.5 ML MTG (1-Thioglycerol,0.45M,1000x)
- 0.5 ML Ascorbic acid (50mg/ml, 1000x)
*Before use, add growth factors into 100ml basal medium with supplements as following:
- 0.1 ML VEGF
- 20 ul FGF-2
- 4 ul BMP-4
Storage Condition
Store 500 ml basal medium at 2-6°C. Store cell growth factors, supplements, fetal bovine serum (FBS) at -20°C. The complete cell culture medium with Supplements can be kept in 4°C for 2-4weeks. Protect from light.
To assure sterility, the prepared medium should be sterilized by filtration through a 0.2 µm filter.
