Description
The Conditioned Medium for Human ESC/iPSC Culture is a complete medium designed for the culture of human embryonic stem cells and induced pluripotent stem cells. Medium contains growth factors and extracellular matrix proteins secreted by MEF feeder cells. Human ES and iPS cells maintain pluripotency when culture in feeder conditions using this medium. It was tested and optimized with human embryonic stem cell (ESC) and induced pluripotent stem cell (iPSC) growth and proliferation in vitro conditioned with mouse embryonic fibroblast (MEF) cells. It is formulated for use with 5% CO2 and 95% air in a humidified incubator. The medium consists of 500 ml of basal medium (containing essential and non-essential amino acids, vitamins, organic and inorganic compounds, hormones, growth factors, trace minerals), supplemented with cell growth supplement, antibiotics, and fetal bovine serum. 15% Fetal Bovine Serum (germline transmission-tested) contains in the medium.
Cell Culture Medium Supplements include:
- 75 ML KOSR (Knockout Serum Replacer)
- 5.0 ML NEAA
- 5.0 ML GlutaMAX Supplement
- 3.5 ul 2-Mercaptoethanol
- 200ul FGF-2 (10ug/ml)
*Before use, add Supplements into 415 ml DMEF/F12
Storage Condition
Store 500 ml basal medium at 2-6°C. Store cell growth factors, supplements, fetal bovine serum (FBS) and antibiotics at -20°C. The complete cell culture medium with Supplements can be kept in 4°C for two months. Protect from light.
To assure sterility, the prepared medium should be sterilized by filtration through a 0.2 µm filter.
